Bromodeoxyuridine (BrdU) labeling is used for birthdat-ing and monitoring cell proliferation. It was originally developed to determine the proliferative index of tumors, and the chemical was used as an antiviral and antineoplastic agent. It was later introduced by Nowakowski et al. to determine the time of origin, migration, lineage, and fate of neuronal cells in the developing nervous system. BrdU labeling is the paradigm the most used to study and characterize, by immunohistochemistry and confocal microscopy, neurogenesis in the adult brain of mammals including in humans. In humans, newly generated neuronal cells, immunopositive for BrdU in the adult dentate gyrus (DG), were identified and characterized in tissue samples obtained postmortem from patients with cancer who had been administered with the drug as part of their therapy. BrdU is not a marker for cell proliferation; it is a marker of deoxyribonucleic acid (DNA) synthesis. Studying and quantifying neurogenesis with BrdU require distinguishing cell proliferation and neurogenesis from other events involving DNA synthesis.
