Aminoacyl-tRNA synthetases (ARSs) are phylogenetically ancient housekeeping enzymes linking amino acids to their specific tRNAs within an ATP-dependent process. The specific recognition of their substrates is critical for the fidelity of protein synthesis and there is normally a single aminoacyl tRNA synthetase for each amino acid, despite the fact that there can be more than one tRNA, and more than one anticodon, for a given amino acid. Among the features of eukaryotic ARSs, there is the distinct property of some of them to form a macromolecular protein complex (multi-tRNA synthetase complex) which is assembled as a group of nine different ARS enzymes plus three non-enzymatic proteins (p43, p38 and p18, known as Aminoacyl-tRNA Synthetase-Interacting Multifunctional Protein, AIMPs). Apparently, complex-forming ARSs are distinguished from non-complex-forming ARSs solely by their ability to interact with the three non-enzymatic factors.
