Confounding influences on phenotype expression in MRL/lpr mice

A colony of 47 four-month-old MRL/MpJ-Fas~(lpr) mice either heterozygous or wildtype for IL10 targeted mutation were reported to the Veterinary Clinical Service because of acute, bilateral, red, ocular discharge. They were scruffy with a diverse array of additional signs. All mice were lethargic, moderately dehydrated, and had multiple subcutaneous swellings on the face and neck. Several mice had multifocal, scaly, epidermal lesions, and the aforementioned discharge from one or both eyes. They were housed on corncob bedding in polycarbonate shoebox-type cages in groups of 4-5 per cage since procurement from Jackson Laboratories. Mice were serologically negative for Ectromelia virus (mouse pox), GDVII (Theiler's) virus, Lymphocytic choriomeningitis (LCM) virus, Mouse hepatitis virus (MHV), Mouse parvovirus (MPV), Mycoplasma pulmonis, Pneumonia virus of mice (PVM), Rotavirus (EDIM), and sendai virus. They were offered water ad libitum from bottles and fed a standard rodent chow diet. The mice were used to investigate the role of autoreactive T cells in the pathogenesis of systemic lupus erythematosus (SLE), which required collection of blood samples from the retro-orbital venous plexus every 4 weeks. Laboratory personnel had recently participated in a mandatory re-training seminar because of technical problems with repeated retro-orbital venipuncture. The colony was not exposed to any deleterious environmental factors or infectious agents. The published life span of MRL/MpJ-Fas~(lpr) mice is approximately 17 to 22 weeks; however, this colony was experiencing unexpected early morbidity and mortality. Unfortunately, the investigator discarded the affected mice before a diagnostic work-up could be performed.
