摘要

The O_2 concentration is an important parameter for tissue homeostasis.Ceils in vitro are usually carried on in atmosphere with CO_2 equal to its amount in the air and in vivo (5 %). Besides, the concentration of oxygen, which is one of the most conservative parameters during cul-turing, corresponds to O_2 content in the air (20 %) that exceeds its physiological value which is about 4 to 7 %. Mesenchymal stem cells (MSCs), that could he very useful as a source of cell material for regenerative medicine, are usually situated in tissues with poor oxygen tension (bone marrow and adipose tissue); The effect of oxygen concentration in the gas phase on the proliferation and osteogenic and adipogenic differentiation of rat MSCs in normoxic and hypoxic conditions has been investigated. We have shown that the proliferation activity of cells under hypoxic conditions (5 % O_2)was higher than those expanded under normoxic conditions (20 % O_2). Besides, induced os-teogenesis and adipogenesis of MSC were better in normoxia than in hypoxia. These findings are important for further applications in regenerative medicine and replacement therapy.
