The product of the NR3C1 gene, the glucocorticoid receptor, plays a crucial role in mediating immune function. The most prevalent translated form in humans is a 777-amino acid protein, which is able to act as a transcription factor, a function that is facilitated by two zinc fingers that constitute a DNA binding domain. Signalling through the receptor is complex and over recent years, elegant experiments to explore alternative splicing to yield GRalpha and beta isoforms by use of two splice acceptors within the ninth exon of GR and the use of variable translation initiation sites have revealed many GR isoforms. These isoforms have been shown to traffic differently to the nucleus and possess a range of transcriptional regulation capacities and target gene specificities [1]. Both alpha- and beta-spliced isoforms may display amino-terminal variability of the translated protein, thus generating a large number of protein isoforms from a single gene, even before the consideration of what effect the numerous known post-translational modifications have upon the function of these proteins.
