It is demonstrated that acceptor stem duplexes derived from native tRNAs which contain athree-nucleotide extension at the Y-terminus of mature tRNA are minimal substrates for ribonuclease pfrom both ESCher-ichia coli and Bacillus subulb-Variants with a cyUdine at position一1are most efficientlyprocessed whereas the G -lvariant represents a comparatively poor substrate.An A-lacceptor stemvanantIs a slightly better substrate than the G-lvariant though generally distinctly less efficient thanthe C一lduplex.This is in qualitative agreement with the frequency of the occurrence of the correspondingnucleotides at position一lin naturalsubstrates,which is highest for pyrimidines and least for G.NMRanalyses of the cozesponding acceptor stems reveal that the confonEation of the nuckotides at position-lcorrelates with the substrate preferences of Ribonuclease P:Whereas C-1adopts a conformationcharacterized by a glycOSidic angle in the anti range (close to high-GMi),the G -1is clearly in synconformation,and that of A-lis intermediate between high-anti and syn-The riboses of nucleotides-lare in all cases predominantly 2'-endo puckered.
