Evaluation of the seed germination of two protected species: Lilium pomponium L. and Lilium martagon L.

The genus Lilium includes near 100 species widespread in Europe, Asia and North America. Among these L. martagon L. and L. pomponium L. are protected by regional laws in Liguria and Piemonte. L. pomponium is a Ligurian-Provencal endemic species growing, in Italy, only in Western Liguria. L. martagon is widely spread in Europe and Asia but, as a result of intensive collection, is rare in Italy. In the genus Lilium seeds germinate by two different types: 'epigeal' or 'hypogeal'. Within each type, the germination can be 'delayed' or 'immediate' related to seed dormancy (Lighty, 1968; Baranova, 1977; Pelkonen, 2005). It is well known that L. martagon, has a hypogeal germination, but nothing is known about L. pomponium. Our experiments allowed to classify the germination of this species as 'delayed epigeal' germination. L. pomponium seed morphology and viability were analysed. Seeds were stored at 15 degrees C with or without dehydration at 12% RH, prior to storage. Germination pre-treatments consisted of chilling (4 degrees C) for 30 or 60 days or warm temperature (24 degrees C) for 30 or 60 days or combination of them. Seeds were then sown in petri with wet filter paper, at 18+or-1 degrees C in dark. The best germination percentage (70%) was obtained after the pre-treatment combination of warm (30 days) and cold (60 days) stratification. The dehydration before storage did not affect germination potential. Fresh L. martagon seeds were used for in vitro germination experiments. A first sterilization trial with NaOCl 4,8% of free chlorine for 3 min, was compared with a treatment with ethanol 70% followed by NaOCl 4,8% of free chlorine for 20 min; both were rinsed several times with sterile distilled water. Seeds were then placed on a solid MS medium with sucrose (3% w/v), and incubated at 20/10 degrees C with a photoperiod of 12/12h (light/dark). After 4 months it was observed that the second sterilization treatment guaranteed higher sterility and that the germination percentage in vitro reached 97%.
