Phytochemical analysis of volatiles from leaves of rugosa rose.

The aromatic compounds and their relative contents of different stage leaves and half opened flowers of five Rosa rugosa cultivars were analyzed by static headspace and gas chromatography-mass spectrometry. The results showed that aromatic compounds do exist in the leaves of R. rugosa. The volatile compounds and their relative contents of leaves were different from each other among the five cultivars. During the leaf development, compounds and their relative contents vary considerably. (1) At roll stage, terpenes were the main aromatic compounds of leaves of four cultivars except for cultivar 'Purple Branch'. The major terpene constituents were germacrene-D and alpha -cubebene in leaves of cultivar 'Saixizi', and alpha -pinene, beta -pinene in leaves of cultivars 'Zifurong', 'Xizi' and 'Puce Dragon'. (2) At expansion stage, there were 20, 25 and 24 kinds of volatiles in leaves of cultivars 'Saixizi', 'Xizi' and 'Puce Dragon', respectively, significantly more than those of roll stage leaves. (3) At mature stage, terpenes were the major constituents, including germacrene-D and 3-carene. (4) The major constituents identified in flowers were alcohols. Phenylethyl alcohol, citronellol and ethanol comprised the principal aromatic compounds for the five cultivars. The flowers also contained alpha -pinene and beta -pinene, but their relative contents were particularly low. Nevertheless, Rugosa rose leaves contained alcohols but no phenylethyl alcohol and citronellol. (5) The relative content of trans- beta -ocimene in roll stage leaves is closely correlated with that of citronellyl acetate in flowers (r=0.8755). This would be useful for breeders to do relative selection at very early stage.
