After a century of empirical vaccine development focused on the “isolate-inactivate-inject” paradigm, vaccinology is moving toward directed development and the rational design of new candidate vaccines [1]. An important direction for vaccinology is that future vaccines may no longer contain live or inactivated whole pathogens. The pneumococcal, meningococcal and Hib polysaccharide conjugate vaccines, the HBV recombinant subunit vaccine and the HPV VLP vaccines are prime examples of new generations of vaccines whose immunogenicity is focused on specific antigenic epitopes. Such subunit vaccines are highly immunogenic, safe, and easier to produce than live whole pathogen biologic products.
