Ubiquitin conjugation has emerged as a key player in the regulation of numerous cellular processes.1 It is best known for its fundamental role in the regulation of protein degradation.2 However, ubi-quitination of lysines on substrates and subsequent binding to the proteasome is not sufficient for effective degradation.3"5 A degradation initiation site has to be presented by the substrate to the proteasome. Intrinsically disordered (ID) regions can act as effective degradation initiation sites. In a seminal study, Prakash et al. have shown that ubiquitinated proteins in vitro are stable unless they contain an ID region.5 Functional ID initiation sites can be located at the protein's termini or internally but, in the latter case, must be much longer.
