In eukaryotic cells, numerous proteins enter and exit the nucleus through the central channel of nuclear pore complexes (NPCs) embedded in the nuclear envelope. This exchange of macromole-cules between the nucleus and the cytoplasm is critical for regulating cell functions and affects most aspects of cellular physiology. More than 20 different nuclear transport receptors mediate nuclear transport pathways in eukaryotic cells. The largest family of nuclear transport receptors is the importin beta (also called karyopherin beta or transportin) family: each member mediates the nuclear import or export of subsets of proteins or RNAs by binding to specific signal peptides or motifs called nuclear localization signals (NLSs) or nuclear export signals (NESs). Although some of the basic mechanisms of transport mediated by importin |3 family have been elucidated, the cargo and signal sequence information specific to each nuclear transport receptor remain to be determined, and one need to have a better understanding of the mechanistic aspect of the regulation of nuclear transport. It is known that transport regulation is achieved at multiple levels: the expression levels and post-translational modifications of each transport cargo, the function of nuclear transport receptors, and the alternation of NPCs.
