Over 60% of the brain dry weight is lipid, with cholesterol being the single most abundant lipid species. Cholesterol is required for instance for myelination [1] and synapto-genesis [2]. Normally, plasmacholesterol is not taken up by the central nervous systenr(CNS) as the blood-brain barrier (BBB) prevents the diffusion of large molecules at the level of tight junctions between brain capillary endo-thelial cells [3]. The ±>ulk of the brain cholesterol is synthesized within the CNS [3] and eliminated across the BBB mainly through conversion into 24(S)-hydroxy-cholesterol [4]. Astrocytes, the most abundant cell type in the CNS, synthesize and secrete cholesterol in apolipo-protein E (apoE) containing lipoprotein particles, and an astrocyte-neuron cholesterol 'shuttle' has been suggested [2], where mature neurons rely on cholesterol produced by astrocytes.
