Fibro/adipogenic progenitors: A double-edged sword in skeletal muscle regeneration.

Adult skeletal muscle is capable of efficient regeneration in response to a variety of insults. Self-renewing myogenic stem cells called satellite cells reside between the myofibres and the basal lamina that ensheathes them. In response to damage, satellite cells proliferate and give rise to myoblasts, which in turn progress through discrete differentiation steps to ultimately fuse with the damaged myofibres or generate new myofibres. However when regeneration fails a fibrotic scar infiltrated with adipocytes (fatty degeneration) ensues. Although the identity of the myogenic progenitors responsible for the majority of regeneration has been clearly elucidated, the cellular origin of fatty infiltration remains controversial. It has been proposed that signals from the damaged environment cue a fate change in satellite cells that are otherwise destined for a myogenic fate. The implication is that the appearance of fibrocytes and adipocytes takes place at the expense of regeneration.
