摘要

Cherry twisted leaf (ChTL)-associated dsRNA was isolated from petioles of symptomatic Bing cherry plants in British Columbia, Canada. The ChTL-dsRNA was isolated by 2 cycles of CF11 column chromatography, T1 RNase and DNase treated, and agarose gelpurified. Oligonucleotide primers based on conserved Closterovirus sequences were used in RT-PCR to generate a ChTL-specific 600 bp fragment. The fragment was digoxigenin labelled and successfully used to detect cloned cDNA produced using dsRNA from theputative ChTL-associated virus in Nicotiana occidentalis. Positive results were also obtained when blots containing total RNA from ChTL-affected Bing cherry, and from N. occidentalis infected with the putative ChTL virus were hybridized with the digoxigenin labelled probe.
