Actively growing shoots on field-grown plants of P. repens cv. Embers were treated with fungicides or growth regulators prior to the collection of multinodal explants (3-5 nodes). The explants were established on half-strength MS medium supplementedwith 1 mg benzyladenine (BA)/litre. Treatment of mother plant shoots with 200 mg BA/litre significantly reduced browning and promoted bud sprouting in vitro. Treatment of mother plants with captafol (Difolatan, 1 g/litre) or iprodione (Rovral Flo, 1 ml/litre) reduced the contamination rate during establishment to 14%, compared with 90% in the control. Bud break in vitro was increased by the addition of gibberellic acid to the medium at 6 mg/litre. Somatic embryos developed from the base of shootlets throughout the multiplication stage.
