Assessment of the microbial status in closed hydroponic system using phospholipid fatty acid analysis.

Phospholipid fatty acid analysis (PLFA) profiles were used to detect the changes in the microbial composition of the rhizosphere of hydroponically grown tomato plants. Root tissues were sampled from two different cultivation systems, i.e. a closed hydroponic deep flow system in a greenhouse and a small-scale climate chamber system. In the closed hydroponic system, tomato (Lycopersicon esculentum) roots were subjected to the following treatments: (i) unbacterized roots (control), (ii) single strainplant growth promoting rhizobacteria (PGPR); Pseudomonas fluorescens 5.014, and (iii) multiple strain PGPR Pseudomonas fluorescens 5.014, Xanthomonas maltophilia [Stenotrophomonas maltophilia] 18.013 and an unidentified Gram-positive isolate 19.018. In the small-scale climate chamber, plants of tomato (cultivars Gitana, Aromata, Armada and Elin), sweet pepper (Capsicum annuum cv. Evident) and lettuce (Lactuca sativa cv. Grand Rapids) were cultivated and harvested after 6 weeks; tomato cv. Gitana plantswere also harvested after 4 and 8 weeks. In addition 6-week-old tomato plants were cultivated in a climate chamber and subjected to the following treatments: (i) control, (ii) Pythium ultimum, (iii) P. ultimum and Pseudomonas fluorescens 5.014 (wild-type), and (iv) Pythium ultimum and Pseudomonas fluorescens5-2/4 (genetically modified type). PLFA profiles, isolated by split-phase extraction and step-wise lipid separation and identified by gas chromatography, revealed significant differences with respectto plant species, cultivars, plant age and treatment with genetically closely related bacterial isolates.
