Asian prunus latent virus: an unusual Potyvirus detected in germplasm from East Asia

Previous work has indicated that certain peach and Prunus mume germplasms from China and Japan were infected by a new Potyvirus designated Asian prunus latent virus (APLV). The virus reacted positively with some plum pox potyvirus (PPV) coat proteinantisera in ELISA, but other tests based on PCR indicated that the virus was not PPV, but possibly a different latent Potyvirus. Data showing some of the unusual features of this virus are presented. Extracts of plant leaves infected with different virus isolates tested positive for the presence of a protein of Mr 68 kD that reacted with antisera to the cylindrical inclusion (CI) protein of tobacco etch potyvirus (TEV) and maize dwarf mosaic potyvirus (MDMV) in western blots. ELISA indicated the presence of a protein that cross reacted with antisera to the CI protein of TEV and MDMV in infected pant tissue. Cytoplasmic inclusions were not detected in thin sections of infected tissue, but were detected in thin sections of PPV or TEV-infected leaves. Immunogold localization failed to reveal the presence of CI proteins in the APLV-infected tissue cells. Antisera to TEV and MDMV coat proteins failed to react with any protein in the infected samples. Only a few flexuous virus particles were found in tissue homogenates subjected to partial purification normally used for potyviruses. It is speculated that APLV is an unusual Potyvirus in that the CI protein translated from the virus genome fails to polymerize into cytoplasmic inclusions characteristic of potyviruses. It is suggested that the virus may have defects that prevent the assembly of virus particles or may have unstable particles which break up during the purification process.
