There is a close relationship between the free proline content (FPC) in Citrus anther and pollen fertility. Proline was extracted with sulfosalicylic acid and determined using the acid ninhydrin method. Significant differences of FPC in anthers existed among cultivars with differing pollen fertility. Cultivars with normal levels of pollen fertility had higher proline contents than those with partial or complete sterility. A positive correlation of FPC existed with percentage pollen germination.
