Dodder transmission of two different MLOs from plum trees affected by 'leptonecrosis'

A graft transmissible disease known as plum leptonecrosis is described. It has been found in Italy since 1933 on Japanese plum (Prunus salicina) and apricot. A phytoplasma is thought to be the causal agent. In Udine, an effort was made to isolate the plum leptonecrosis agent (PLNa) from plum and peach to Catharanthus roseus. A clone of Cuscuta campestris was used as a vector. Six different types of inoculum sources were used, including young shoots of Myrobalan (Prunus cerasifera), 2-year-old plants of Ozark Premier, micropropagated Shiro plants, potted seedlings of peach GF 305 infected by chip-budding from plum, cut Ozark Premier shoots, field infected plum trees. Of 176 C. roseus plants inoculated, 9 became infected; 4 reacted with typical small pale flowers and 5 with virescence. In sub-inoculated C. roseus, the symptom types remained invariable. Both sources of inoculum and test plants were observed under EM and light fluorescence microscope. The results indicated that plum can host different phytoplasmas, at least 2 in the investigated area. The aetiological involvement of the 2 different phytoplasmas is discussed as well as the possible occurrence of mixed infections. Analysis with PCR of naturally and artifically infected plants are in progress.
