PCR has recently been used in Spain to detect phytoplasmas associated with fruit tree diseases, such as apricot chlorotic leaf roll, apple proliferation and pear decline. For a PCR reaction, a fragment of the 165 rRNA gene was selected as template DNA and specific and non-specific primers were used. PCR amplification products were analysed by RFLP with AluI and RsaI enzymes. Preliminary results were obtained for the presence of phytoplasmas in fruit trees, as was previously described for apricot, plum and apple trees. This is the first report of the presence of phytoplasmas in pear trees in Spain.
