Rooting percentage of Asparagus cv. Tainan Selection 3 (TS 3) nodal sections in MS medium with NAA, kinetin and 3% sucrose was previously found to be very low (1.7%). In this study, tryptone (250 mg/litre), phloroglucinol (PG; 162 mg/litre) and ancymidol (1.28 mg/litre) were added to a modified MS medium containing NAA (0.15 mg/litre), kinetin (0.15 mg/litre) and 6% sucrose to investigate their potential for increased in vitro rooting of TS 3. The rooting percentage in control medium was 18%, while in media with tryptone, PG or ancymidol, the rooting percentages increased to 41, 59 and 63%, respectively. With a combination of tryptone or PG with ancymidol, in vitro rooting reached 73-78%. In addition to increasing the frequency of rooting, the presence of PG or tryptone significantly increased shoot number and root length. In contrast, ancymidol reduced shoot length, but significantly increased the diameter of shoots and roots and number of shoots per plantlet. Survival rate oftransplanted plantlets was influenced by temperature, covering with a plastic bag and inoculation with Glomus spp. to induce vesicular-arbuscular mycorrhizal associations.
