Chloroplast DNA variation in sour cherry

Restriction fragments of chloroplast DNA (ctDNA) from 29 sweet (Prunus avium), sour (P. cerasus), and ground (P. fruticosa) cherry selections and species hybrids were analysed to determine the maternal origin of the tetraploid sour cherry. Total leaf DNA was isolated and digested with two restriction endonucleases that preferentially cut ctDNA. The DNA fragments were then separated on agarose gels and filter-hybridized with two PstI fragments of tomato ctDNA. RFLPs were identified that differentiated the ctDNA of sweet and ground cherry. Of the 20 sour cherry selections, 17 had the ctDNA RFLPs of ground cherry while 3 had the ctDNA RFLPs of sweet cherry. These results indicate that the allotetraploid sour cherry arose independently at least twiceand that the more common maternal species is ground cherry.
