Isolation and characterization of apple chlorotic leaf spot trichovirus (ACLSV) strains occurring in fruit trees in Poland, preparation of antisera and elaborating methods for routine ACLSV diagnosis in fruit trees were studied. ACLSV was detected in apple, pear, sweet cherry, plum and quince, using indirect F(ab')2 ELISA. Six isolates of ACLSV obtained from apple, pear, plum and sweet cherry were studied and antisera were prepared against 5. Plum isolate SX/2 showed a significantly weaker reactionin ELISA with 2 antisera (prepared against apple and cherry isolates) than the other isolates. Isolate JPom, obtained from apple, reacted better with monoclonal antibodies EMA32 than the other isolates. The coat protein (CP) of the SX/2 isolate migratedfaster in SDS-PAGE than CP of other isolates tested. RT-PCR was applied to the isolates using samples of herbaceous and woody hosts. Specific products were obtained for all the isolates, except SX/2, and digested with 3 restriction enzymes. Two different patterns were found for 4 isolates.
