Genetic diversity within and among P. lunatus var. silvester populations from the central valley of Costa Rica was evaluated using enzymes and microsatellite markers. Microsatellite markers detected a greater level of genetic diversity than the enzyme markers. However, the relatively moderate genetic diversity can be explained by restricted origin of samples or by the source of the primer. Total genetic diversity and within population genetic diversity were underestimated with enzyme markers. The contribution of genetic diversity among populations to total genetic diversity was similar for both markers. Enzymes and microsatellite markers revealed a high inbreeding level for whole population and within population (FIS). Within-population genetic diversity accounted for 70% of the genetic diversity detected by both genetic markers. Lima bean wild populations at the central valley of Costa Rica may constitute a valid in situ conservation unit..
