I read with interest the recent article by Yik et al (1), in which they reported that cyclin-dependent kinase 9 (CDK-9) inhibition protected against a primary inflammatory response in cartilage. According to the authors, CDK-9 activity was suppressed by the pharmacologic CDK-9 inhibitor flavopiridol, and flavopiridol suppressed the induction of various inflammatory mediator genes by interleukin-1/3 (IL-1/3) as well as the induction of catabolic genes. In contrast, it had no effect on the expression of anabolic genes and housekeeping genes. One method used to achieve these results was polymerase chain reaction (PCR) array analysis. However, some of the results obtained using this approach appear to be contradictory.
