A simple and effective method for evaluating hundreds of soybean genotypes is very useful for salt tolerance genetic studies and breeding. Afield evaluation can handle a large number of materials. However, the salt distribution has gradient either vertical or horizontal in a salinity field. This may result in an incorrect evaluation. Hydroponics can provide a reliable evaluation, however it is labor and cost consuming. In this report, we described a salt-water flooding method for evaluating salt tolerance for a large number of plant materials. When this method was applied to screening of 174 wild soybean (Glycine soja) accessions from Japan, several genotypes, such as JP30932 and JP30942, were identified to be salt tolerant. Evaluation of these soybean accessions using hydroponic method with a half strength of Hoagland and Arnon nutrient solution containing 100 mM NaCl gave a similar evaluation result as the salt-water flooding method did, indicating that the salt-water flooding method was simple and effective for screening large number of soybean genotypes, and it also had potential use in other crops. The salt tolerant wild soybean accessions, JP30932 and JP30942, might be used as genetic resources for improvement of salt tolerance in cultivated soybean.
