We evaluated the performance of three quantitative assays for antibody to hepatitis C virus (HCV),Cobas core anti-HCV-EIA (assay C), Immucheck F HCV-Ab (assay I),and Lumipulse II Orth HCV (assay L) in serial samples from 454 patients of Tokyo Metropolitan Komagome Hospital.Quantitative assays were compared with assays for anti-HCV core antibodies using core antigens JCC-2 and Cll, and the relationships between HCV antibody titers expressed by cut-off index and HCV RNA positivity by polymerase chain reaction were analyzed.Overall concordance of anti-HCV antibody positivity between assays C,I and L was a high 97.4%.There were significant correlations between the results of anti-HCV core antibody assays and both assays C and I, but not assay L. With regard to patients who had not received interferon, in HCV RNA-negative sera,anti-HCV titers (mean+-D) measured by assay C, I or L were 93.7+-91.7, 21.1+-30.1 and 4.37+-4.18, respectively, while values in HCV RNA-positive sera were 753.4+-430.1,236.3+-181.4 and 94.2+-101.4,respectively.Therefore, the existence of HCV RNA may be estimated from anti-HCV titers as measured by these quantitative assays.In conclusion, these automated quantitative assays proved to be not only a convenient test for the titration of antibody to HCV but also useful in the estimation of HCV viremia.
