Crossreactivity Between Curvularia lunata and Alternaria alternata

Background: Several fungi, including Alternaria alternata and Curvularia lunata, induce respiratory allergy disorders in atopic individuals. Fungal proteins show crossreactivity and can facilitate the selection of relevant allergens. We studied crossreactivity between A. alternata and C. lunata proteins using hypersensitive patients' sera, and its implications for improving immunotherapy for fungal allergies. Methods/Data base: A. alternata-hypersensitive patients (n = 20) were selected for the present study. IgE binding in sera was studied by enzyme-linked immunosorbent assay (ELISA), immunoblot, ELISA inhibition, and immunoblot inhibition with C. lunata and A. alternata extracts. Skin tests were carried out with C. lunata and A. alternata extracts on 253 patients with respiratory allergy. Results: ELISA using Curvularia and Alternaria as a solid phase antigen with Alternaria-hypersensitive sera demonstrated a positive correlation for allergenicity (p = 0.030). Skin tests results of 253 patients with Alternaria and/or Curvularia demonstrated a significant correlation (p = 0.0001). Immunoblot with Alternaria-sensitive individual patients' sera identified 9-13 immunoglobulin E (IgE) binding proteins in Alternaria extract and 7-13 IgE-binding proteins in Curvularia extract. ELISA inhibition with individual patients' sera showed a close relationship between these extracts. Alternaria-sensitive patients' sera showed IgE binding with a major allergen Cur 1 1, serine protease from Curvularia. Immunoblot inhibition showed that proteins of 12, 23, 26, 31, 41, 45, 65, and 75 kDa were common in both extracts. Conclusion: Significant crossreactivity was demonstrated between Curvularia and Alternaria extracts and hence either of the extracts will serve for detection of Alternaria or Curvularia allergy.
