Refining humane end points in mouse irradiation studies

摘要

With increasing interest in stem cells and gene function in cell biology, there has been an increase in experimental protocols which involve the whole body irradiation of mice. In these techniques the recipient mouse is exposed to a dose of radiationthat destroys its bone marrow before receiving a transplant of bone marrow progenitor cells from a donor animal or embryo. If the transplant is successful the progenitor BM cells from the donor animal will establish a new haematopoietic system in the recipient mouse. The development of the transplanted bone marrow can be followed by serial blood sampling or bone marrow biopsy. It is important that enough of the host animal's bone marrow is destroyed to allow the transplanted cells to implant and developin the vacant niche. If the recipient mouse was not "rescued" by the BM transplant then it would die from failure of its bone marrow as a result of exposure to the radiation.
