Thirty-four years ago. two years after the first high-resolution X-ray structure analysis of a protein (myoglobin) by Kendrew, crystals of cytochrome c oxidase (COX) from bovine heart were obtained by Yonetani. But all efforts to determine the three-dimensional structure of the enzyme at the atomic level failed due to the poor quality of crystals. Now high-resolution (2.8 A) X-ray crystal structures of the oxidized COX from Para-coccus denitrificans and from bovine heart have been published almost simultaneously by Michel and co-workers. and by Yoshikawa and co-workers. respectively. In the two cases, however, different approaches were used. Michel et al. obtained crystals of COX from Paracoccus only after forming a complex with the variable fragment of a monoclonal antibody, whereas Yoshikawa et al. obtained crystals of COX from bovine heart only after addition of the detergent decylmaltoside during crystallization.
