A retrospective study of well characterized isolates of Clostridium perfringens strains from 10 food poisoning outbreaks was performed in order to determine epidemiological links and to evaluate molecular methods. The strains were collected over a period of nearly one decade (1984 to 1991) in south-east Germany. In total 35 strains, 12 of them isolated from food and 23 from patients, which consumed the respective food, have been investigated. Methods applied were the analysis of the total soluble cytoplasmatic proteins by proteinfingerprinting and pulsed-field gel electrophoresis (PFGE) with four different macro-restrictionenzymes (Smal, Kspl, Apal, Sa/l). PFGE showed with all enzymes similar results. in nine outbreaks PFGE was able to confirm, that the food and the patients isolates were identical. The enzyme Sail was most cost effective. Protein analysis could be interpreted only with difficulties and was not as reliable as PFGE. The methods under investigation, especially PFGE, proved to be useful for science-based examinations of clostridia in food-borne diseases in man. For routine examinations they are of minor interest because they are more expensive and laborious than conventional methods.
