Application of principal component analysis for data process in metabonomic studies

Aim To investigate biochemical mechanism of treating hyperactivity syndrome with Complex Ucncaria-stem mixture (USM). Methods In mouse model with apomorphine, the biochemical mechanism was studied by metabonomics method to evaluate the ability of drug to improve and regulate neurotransmitter through the determination of three kinds of monoamine neurotransmitters dopamine (DA), noeradrenaline (NE) and 5-hydroxytryptamine (5-HT) in both brain and plasma by HPLC-ECD method. Principal component analysis (PCA) was performed using multivariate statistics package in Matlab 7.0.1 R14 SP1 (Math Works). Results Complex Ucncaria-stem decoction can increase plasma DA concentration to the level close to that of control group and it can reduce DA concentration in brain to the level close to that of control group. The results showed that USM could increase DA concentration; reduce 5-HT concentration in plasma and DA concentration in mouse brain tissue. All these indicated that USM could regulate the unbalance of monoamine neurotransmitter, thus treat hyperactivity syndrome. Conclusion All these indicated that the therapeutic effect of USM on hyperactivity syndrome was related to the biochemicals base on which it regulated the unbalance of monoamine neurotransmitter. PCA result suggests that the metabonomics method may play an important role in identifying markers for producing the therapy action and funding the action mechanism of the traditional Chinese medicines to be related with down-regulating DA and 5-HT levels in brain tissue.
