The present study indicated that the addition of fluoride did not inhibit In vivo nitrate reductase activity in leaves of dark grown seedlings, while there was tremendous inhibition in leaves of light grown seedlings. Fluoride had no significant effect on in vitro nitrate reductase activity. Inhibition In leaves of light grown seedling in in vivo nitrate reductase activity increased with the enhanced concentration of fluoride in assay medium. The inhibition in in vivo nitrate reductase activity exerted by the fluoride in leaves of light grown seedlings was partially reversed by the addition of pyruvate, succinate and malate in assay mixture. The result indicated that NADH generated in the reaction occurring beyond the triosephosphate dehydrogenase step of glycolysis may be utilized in nitrate reduction in light. Experiment with leaves of dark grown seedlings showed that addition of glycolytic intermediates in assay medium did not inhibit the nitrate reductase enzyme activity. This suggested that the source of NADH in dark grown seedlings is from glycolytic pathway. In conclusion, the observed effect of fluoride on leaves of light and dark grown seedlings of wheat depended on the physiological availability of NADH.
