RNA interference is an evolutionarily conserved gene-silencing pathway in which the nuclease Dicer cleaves double-stranded RNA into small interfering RNAs (Hannon, 2002). The Dicer gene encodes a protein possessing an RNA helicase motif containing a DEXH box in its amino terminus and an RNase motif in the carboxy terminus (Matsuda et al., 2000). The encoded protein functions as a ri-bonuclease and is required by the RNA interference and small temporal RNA pathways to produce the active small RNA component that represses gene expression (Hutvagner et al., 2001; Ketting et al., 2001; Knight and Bass, 2001). In higher vertebrate cells, Dicer-related RNA interference-is involved in the formation of the heterochromatin structure. A conditional loss-of-function mutant of Dicer in chicken DT40 cells causes defects in heterochromatin formation and chromosome segregation. Depletion of Dicer results in cell death with the accumulation of abnormal mitotic cells that show premature sister chromatid separation. Aberrant accumulation of the transcript from or-satellite sequences, which consist of human centromeric repeat DNAs, was detected in Dicer-deficient cells (Fukagawa et al., 2004; White and Allshire, 2004). In spite of our knowledge of the biological function of Dicer, its functional and hereditary mechanism in chicken is not fully understood. In this report, precise localization of Dicer was first performed by RH mapping in chicken. We also compared the coding region of Dicer in different species.
