One of the hallmarks of the fish immune response to a foreign protein is the production of antibodies which bind specifically to epitopes of the target molecule. Because that antibody response is the product of interactions between several components of the immune system, quantification of the antibody response to a defined protein provides an attractive approach for examining changes in the humoral arm of the fish disease defences. Measures of the antibody response have been used to help describe the impact of environmental conditions, physiological changes, vaccination, and pathogenic micro-organisms on fish health. Except for vaccine trials and research studies using particular fish pathogens, antigens used in fish studies are often chosen on the basis of their immunogenicity in mammals. The fish antibody response, however, is generally weak compared to the IgG response of mammals. Few laboratory studies have focused on determining which antigens will elicit a strong antibody response in fish. Hodgins et al. examined the immune response of rainbow trout (Oncorhynchus mykiss) to several protein antigens, but the investigators were limited to measuring the specific antibody levels by immunodiffusion and the haemagglutination test. In the current study, a variety of protein antigens were administered to fish under standardised conditions and the resulting antibody responses were compared on the basis of an enzyme-linked immunosorbent assay (ELISA). The goal of this study was to identify one or more immunogenic protein antigens that would be suitable for characterising the effects of various environmental conditions on the humoral response of salmonids. Whenever possible, inexpensive, commercially prepared protein antigens were selected for testing to ensure that each would be readily available to most fish health research laboratories, and of uniform composition.
