When I joined the laboratory of Frederick J. de Serres in the Biology Division of the Oak Ridge National Laboratory as a Postdoctoral Fellow in 1960, he was developing the adenine-3 (ad-3) for-ward-mutation test in Neurospora crassa. This test utilizes a direct method based on the accumulation of purple pigment by ad-3 mutants, rather than their requirement for adenine [1]. Control or treated conidia are incubated in 12-1 Florence flasks containing 10 1 of minimal medium supplemented with sorbose, sucrose, and a low concentration of adenine. After 7 days of incubation with aeration in the dark at 30 deg C, the purple (ad-3 mutant) colonies contrast with the background of white (wild-type) colonies. I had proposed to use this intriguing new test to determine the mutagenicity of certain base analogs, especially 2-aminopurine, in N. crassa. As a graduate student (1956-1960), I had read with great interest (and probably without much understanding) the papers of Benzer and Freese on the various aspects of the mutagenicity of base analogs, such as 2-aminopurine, in the rII region of bacteriophage T4 [2-5]. Later, I came to recognize these papers as classics among the early papers on proposed molecular mechanisms of mutation, and I have long included them in my genetics courses.
