The 4th International NKT Cell & CD1 Workshop was held 4-8 October 2006 at the Abbazia di Spineto, in the heart of Tuscany. This report highlights cutting-edge research and issues discussed at the meeting. Studies of lipid-specific T cells have identified many facets of their biology. Some features resemble those of peptide-specific T cells, whereas others are distinct and reflect the biochemical nature of recognized antigens. Like peptide-specific T cells, lipid-specific T cells also develop in the thymus and require the appropriate presentation of endogenous antigens, although the molecular mechanisms controlling their development are different. Lipid-specific T cells also recognize microbial antigens and participate in immune protection during infection, in autoimmuniry and in tumor surveillance and also direct T helper type 1 (T_H1) and T_H2 polarization. Finally, a chief population of lipid-specific T cells, which express markers of natural killer (NK) cells and therefore are known as NKT cells, has important regulatory functions that may dampen or exacerbate inflammatory responses. These issues were discussed at the '4th International NKT Cell & CD1 Workshop' at the Abbazia di Spineto in Tuscany.
