A role for transcription factor Myb in breast cancer (BC) has been considered for decades, with substantive data showing an association with estrogen receptor a (ERalpha)-positive BC. Recently, this has been moved into a functional milieu, where Myb-dependent proliferation and impeded differentiation have been established as well as progression in vivo3 and that MYB transcriptional elongation is mediated by ERalpha in an estradiol-dependent manner. High Myb in ERalpha+ BC predicts excellent survival, and some have suggested that MYB is a tumor suppressor gene. Another way to think about this is that this group of BC is very effectively treated. Nevertheless, it is clear that other forms of BC are not so effectively treated yet Myb is still expressed. Whether Myb is important in these is less clear. Central to this lack of clarity is defining the role of Myb in metastasis
