Understanding the molecular mechanisms that underlie the fate choices of somatic stem cells is a major goal of today's research as this is essential for the potential exploitation of stem cells in regenerative medicine. Moreover, this knowledge is of great significance when it comes to counteracting the functions of cancer stem cells (CSCs), which are believed to be regulated by mechanisms similar to those in normal stem cells. Increasing evidence points towards a pivotal role of epigenetic mechanisms in orchestrating the highly complex gene networks which guide the options of a stem cell along multiple possible fates. It has been known for quite some time that differential histone tail modifications can control stem cell functions. However, only very recently it has been appreciated that DNA methylation represents another dynamic epigenetic mark that undergoes extensive remodeling during the differentiation of self-renewing stem cells.
