Eph receptors constitute the largest family of tyrosine kinase receptors. They interact at direct cell-cell contacts with their membrane-bound ephrin ligands, which also are capable of signaling. Ephrins and Ephs regulate many diverse processes during embryonic development, in most cases by affecting the cytoskeleton.In spite of the notoriety of tyrosine kinase receptors as regulators of cell proliferation, it was initially thought that Eph receptors represented an exception and that they were unable to affect the cell cycle. More recently, Ephs and ephrins have been found to be expressed in several adult stem cell niches, such as the brain, skin and intestine, and to regulate the proliferation of stem/progenitor cells. Moreover, Eph receptors can act both as tumor promoters and tumor suppressors, depending on the cellular context.
