Members of the structural maintenance of chromosome (SMC) family of proteins form specialized molecular machines that perform vital functions in chromosome biology. The characteristic ring-shaped structure of the SMC complexes, together with their ATPase activity, makes them uniquely suited to regulate the dynamic structure of chromosomes. SMC complexes are best known for their roles in chromosome condensation and sister chromatid cohesion, which are mediated by the Smc1/3 and Smc2/4 complexes, respectively. A third and less understood SMC complex involving the Smc5 and Smc6 proteins is intimately involved in DNA damage repair (reviewed in refs. 1 and 2). Like the other SMC complexes, the Smc5/6 complex is required for cell viability and is highly conserved among eukaryotes. The Smc5/6 complex contains six additional Non-SMC Elements (Nse1-6), including the Mms21 (Nse2) sumo-ligase. Genetic and biochemical studies of hypomorphic smc5, smc6 and mms21 alleles point to the direct involvement of the Smc5/6 complex in repair via homologous recombination (HR). Cells expressing the sumo ligase-deficient allele of Mms21, mms21-CH, were shown to accumulate X-shaped DNA structures at stalled replication forks.
