Quality control in oocytes: domain-domain interactions regulate the activity of p63.

Understanding the evolution of tumor suppressor proteins has turned out to be difficult based on the observation that most tumors develop late during the lifetime, when the reproductive period has already ceased. This observation has suggested that the original function of tumor suppressor proteins must have been different and more related to basic cellular processes. At least for the p53 protein family, this connection to more basic functions in the development of cells and in metabolism seems to be firmly established. New insight into the potential evolution of p53 has also been obtained from investigations of the two other mammalian family members, p73 and, in particular, p63. Knockout mouse studies of p63 have demonstrated its role in maintaining epithelial stem cells. In addition, a second role of p63 as a quality control factor in oocytes was identified. The discovery of family members in invertebrates such as C. elegans that do not develop cancer but that use their p53 family members for the surveillance of the genetic stability of germ cells suggests that quality control in germ cells is one of the original functions of the entire protein family.
