Lung cancer is the most common cause of cancer-related death in the United States. In solid tumors including lung cancer, tumor cells proliferate at a rate that exceeds the oxygen supply, resulting in regions of low oxygen tension (hypoxia). Tumor cells adapt to hypoxia by inducing genes involved in angiogenesis or glucose metabolism via the transcription factor hypoxia-inducible factor (HIF). HIF consists of two subunits, HIF-alpha and HIF-beta. Adaptation to hypoxia increases tumor cell invasiveness and metastatic potential, which are the main causes of cancer death. Although HIF is generally considered to promote cancer progression and many efforts have been made to screen for HIF-1 inhibitors as attractive therapeutic agents for cancer, a recent report suggests that deletion of HIF-2a promotes lung cancer development. Thus, the role of HIF in lung cancer is complex and may depend on the stage of cancer development.
