摘要

Canonical Wnt signaling plays a fundamental role in multiple biological processes, including the establishment of the early anterior-posterior body axis and dorsal-ventral patterning of the nervous system. Misregulation of Wnt signaling is frequently associated with human disease-most prominently cancer. The canonical Wnt pathway is activated by the binding of a Wnt ligand to its receptors at the cell membrane, which leads to the translocation of beta-catenin into the nucleus, where it interacts with Tcf/Lef transcription factors through their N-terminal domains. This interaction converts the Tcf/Lef proteins from repressors into activators (Fig. 1) and results in activation of their target genes.
