Checkpoint kinase-1 (CHK1) is well known for its role as the effector kinase in the ATR-CHK1-CDC25 DNA damage response, a key pathway in maintaining genomic integrity. CHK1 is activated by the "ATM and Rad3-related" (ATR) kinase in response to stalled replication forks, bulky lesions and single-strand DNA breaks. Activated CHK1 then inactivates CDC25, the protein phosphatase that converts cyclin-dependent kinase-1 (CDK1) into its active form. Thus, CHK1 prevents CDK1 from driving cells with damaged or incompletely replicated DNA into mitosis. However, CHK1 is essential for cell viability, and suppression of CHK1 activity can result in unscheduled DNA replication that induces DNA damage, suggesting that CHK1 is also active during normal mitotic cell cycles.
