Estimation of glucosinolates in rapeseed - a review

Among the antinutritional factors present in rapeseed, glucosinolates are the ones in which scientists are more interested than others due to their toxic and goitrogenic effects. Various methods have been developed from time to time to estimate glucosinolates in rapeseed, ranging from simple test paper, enzymatic, volumetric methods to complex HPLC, ELISA, chromatographic methods. But most of the methods developed so far suffer from one or another drawback. Even today, a need exists to develop a simple, rapid, accurate, economical and reliable method. These methods are mainly classified into two types: i) Estimation of individual glucosinolates - which are time-consuming but give qualitative as well as quantitative measurement of glucosinolates; and ii) Estimation of total glucosinolates - which are rapid and advantageous when large numbers of samples are to be analysed for screening purposes. It gives only quantitative data. Important steps of Analysis of glucosinolates are i) Extraction of glucosinolates; ii) Removal of interfering compounds; and iii) Analysis of glucosinolates. Gas chromatography (GC) and high performance liquid chromatography (HPLC) are the most widely used methods. Lack of pure reference glucosinolates is also one of the major difficulties facing any analyst. In the present context, an attempt has been made to review various methods developed and modified till now for estimation of glucosinolates in rapeseed along with their efficiency and drawbacks. Analytical methodologyfor glucosinolates is continuing to evolve and improve and undoubtedly new methods will appear which will further facilitate analysis.
