A variety of chemistries and purification approaches can be used to manufacture synthetic oligoribonucleotides for use in RNA interference (RNAi). All of these methods can result in high quality oligoribonucleotides, however scale up to high throughput or large scale synthesis can be challenging. A description of quality control issues from a manufacturing perspective will be discussed using the synthesis of Dicer-substrate small interfering RfVAs (DsiRNAs) as an example.
