This report describes the actions of soman on brain in the rat experimental animal model and the effectiveness of eight different antidotal pretreatments. The behavior, local cerebral glucose use (LCGU), brain muscarinic receptor and histology are described for the seizure phase (1 hr. after soman), neuropathology phase (72 hr after soman) and recovery phase (10 days after soman). Antidotes studied were representatives from five different classes: muscarinic receptor blockers, nicotinic receptor blocker, reversible inhibitors of acetylcholine esterase, cholinesterase reactivator, anticonvulsant. The report also compares soman with other convulsants (kainic acid, bicuculline) and with the anticholinesterase, diisopropylphosphorofluoridate (DFP). Calcium channel blockers given intracerebrally did not provide reliable protection against the seizure induced pathology. Although some of the antidotes provide important protection, they fall short of the desired goal. The limited protection provided by the traditional antidotes studied in this project emphasizes the need for new ideas. Since there are endogenous substances which can inactivate soman or could counter its actions, we suggest that research along those lines could be promising. (aw)
