In this thesis scanning tunneling microscopy (STM) and atomic force microscopy(AFM) techniques are applied for surface characterization. A home-built STM was first used for investigating different substrate materials in order to find the most suitable ones for special STM studies. Secondly, STM was used to get new information about the surface structure of protein bacteriorhodopsin, of purple membrane in added soyaphosphatidylcholine matrix Langmuir-Bodgett film. The
