摘要

Thermus sp. E86264 was cured of plasmids by the RNA polymerase inhibitor, acridine orange, and a plasmid was isolated and characterized. Plasmid profiles were analyzed by electrophoresis and a physical map of the plasmid was constructed using five site-specific endonucleases; Hind III, Pvu II, Nco I, Sal I and Dra III. Single restriction sites for Nco I, Sal I, and Dra III respectively were present. The size of the plasmid was 11kb, and the plasmid encoded for protease activity, and was associated with trimethoprim resistance. The size and the single restriction sites indicate that this plasmid, pTE264-1, may represent a useful cloning vector in Thermus and other thermophiles.
